چکیده

Multiple myeloma is a plasma cell cancer with poor survival, characterized by the clonal expansion of multiple myeloma cells (MMC), primarily in the bone marrow. Novel compounds are currently tested in this disease, but partial or minor patients’ responses are observed for most compounds used as a single agent. The design of predictors for drug efficacy could bemost useful to better understand basic mechanisms targeted by these drugs and design clinical trials. In the current study, we report the building of a DNAmethylation score (DM score) predicting the efficacy of decitabine, an inhibitor of DNA methyltransferase (DNMT), targeting methylation-regulated gene expression. DM score was built by identifying 47 genes regulated by decitabine in human myeloma cell lines and the expression of which in primary MMCs of previously untreated patients is predictive for overall survival. A high DM score predicts patients’ poor survival, and, of major interest, high sensitivity of primaryMMCsor humanmyeloma cell lines todecitabine in vitro. Thus,DMscore could beuseful to design novel treatments with DMNT inhibitor in multiple myeloma and has highlighted 47 genes, the gene products ofwhich could be important formultiplemyelomadisease development.MolCancer Ther; 11(12); 1–8.

Utilization of medium-chain triglycerides by neonatal pigs: effects of emulsification and dose delivered.

Four trials were conducted using 86, 24-h-old pigs to evaluate the utilization of medium-chain triglycerides (MCT). Effects of emulsification and amount of MCT fed were examined. After a 4-h period during which feed was withheld, pigs were force-fed MCT (containing 75% octanoate and 25% decanoate), marking time 0 of the experiment. Blood samples were obtained at 1 and 2 h for subsequent medium-...

HIV-1 tropism evolution after short-term maraviroc monotherapy in HIV-1-infected patients.

We analyzed the evolution of viral tropism after 8 days of maraviroc monotherapy, i.e., we used the maraviroc clinical test (MCT), in 21 patients with and 14 without virological response to the drug (MCT(+) and MCT(-) patients, respectively). No increases in CXCR4 inferred viral loads (X4IVLs) were observed in MCT(+) patients, while X4IVLs increased only in MCT(-) patients, with X4IVLs of >2 lo...

Effect of exercise and medium-chain fatty acids on postprandial lipemia.

The purpose of this study was to evaluate the effect of medium-chain triglycerides (MCT) with and without exercise on postprandial lipemia (PPL). Subjects were 25 young men and women. Each subject performed three trials: 1) control (fat meal only, 1.5 g fat/kg) 2) MCT (substitution of MCT oil, 30% of fat calories), and 3) MCT + Ex (exercise 12 h before the MCT meal). Before each trial, the subj...
